ABSTRACT

Abstract Acmella uliginosa, an edible herb belonging to Asteraceae family, was collected from the Terai region of Uttarakhand, India. Methanol and hexane extracts of the whole plant were prepared using soxhlet apparatus. The GC-MS analysis of plant extracts identifies 22 and 35 major compounds of methanol and hexane extracts which comprises of 74.21% and 73.20% of the total composition of extracts, respectively. The major compound in hexane was 2, 4-heptadienal (7.99%) whereas trans, trans-9, 12-octadecadienoic acid propyl ester (16.96%) was major compound in methanol extract. The extracts were evaluated for antioxidant and anti-inflammatory properties. Methanol extract showed higher free radical scavenging and reducing power activities with IC50 value 153.82±1.69 µg/mL and RP50 value of 152.28±0.41 µg/mL, respectively. The metal chelating activity was higher in hexane extract as compared to methanol extract i.e., 62.08±0.25 µg/mL. The anti-inflammatory activity assessed by its ability to inhibit denaturation was higher in methanol having IB50 value 87.33±0.15 µg/mL. The total phenolic content (TPC), total flavonoid content (TFC) and ortho-dihydric phenol content (ODP) of methanol and hexane extracts were also evaluated. TPC, TFC and ODP was higher in methanol extract having value of 122.23±0.22, 35.01±0.29 and 8±0.86 mg/mL, respectively. Acmella uliginosa, might be considered as a natural source for antioxidant and anti-inflammatory properties

ABSTRACT

Abstract Curcumin, contained at Turmeric (Curcumalonga), can exert many beneficial pleiotropic activities in the gastrointestinal tract. This study evaluated the antioxidant and anti-inflammatory activity of C. longa on 5-fluorouracil (5-FU)-induced oral mucositis (OM) in hamsters. Phytochemical analysis of crude C. longa extract (CLE) was performed to detect the presence of curcumin by TLC and HPLC. Golden Syrian hamsters were orally pre-treated with CLE (5, 50, or 100mg/kg). Cheek pouch samples were subjected to macroscopic and histopathological evaluation. ELISA was performed to quantify the inflammatory cytokines IL-1ß and TNF-α. Superoxide dismutase (SOD), glutathione (GSH) and malondialdehyde (MDA) levels were assessed by ultraviolet-visible spectroscopy analysis. Behavior analysis was conducted by the open field test. Curcumin content in the CLE was 0.55%m/m ± 0.0161 (2.84%). The group treated with 5mg/kg CLE showed healing evidence with macroscopic absence of ulceration (p<0.05) and microscopic aspect of re-epithelialization, discrete inflammatory infiltrate and absence of edema. Treatment with 5mg/kg CLE significantly increased GSH levels, and reduced MDA levels and SOD activity (p˂0.05), and decreased IL-1ß (p˂0.05) and TNF-α (p˂0.01) levels. A significant reduction in walking distance, ambulation, speed, and rearing was observed for motor activity. Curcumin reduced oxidative stress, inflammation, and motor activity in hamsters with 5-FU-induced OM.

Abstract In this study, it was aimed to investigate the amount of antioxidant, protective properties against DNA damage and antibacterial properties against various pathogens after the interaction of Ag metal (Ag NPs/Sa) of Sophora alopecuroides L. (S. alopecuroides L) plant seed, which is grown in Igdir and used in the treatment of many diseases. The DPPH radical quenching activity of Ag NPs/Sa was performed by using Blois method, DNA damage prevention activity by gel electrophoresis and antibacterial property by disk diffusion method. With the green synthesis method, AgNPs obtained as a result of the reaction of the plant and Ag metal are UV visible spectrophotometer (UV-vis), fourier-transformed infrared spectroscopy (FT-IR), X-ray diffraction (XRD) and scanning electron microscope (SEM). DPPH radical quenching activity of Ag NPs/Sa was investigated in the concentration range of 25-250 µg/ml. The radical quenching activity at a concentration of 250 µg/ml was 85,215 ± 0,101%, while this value was 93,018% for the positive control BHA. It has been observed that the protective property of pBR322 plasmid DNA damage against OH radicals originating from H2O2 increases with concentration. It has been observed that Ag NPs/Sa has significant antimicrobial properties against some pathogens (B. subtilis ATCC 6633 E. coli ATCC 25952, B. cereus ATCC 10876, P. aeruginosa ATCC 27853, E. faecalis ATCC 29212, S. aureus ATTC 29213 and C. albicans ATTC 90028) that cause disease and even some pathogens are more effective than antibiotics

ABSTRACT

Abstract Epidemiological studies suggest that acute kidney injury has certain effect on myocardial function. In this study, for the first time, we tested a boron compound namely lithium tetraborate an act as an anti-oxidant and anti-inflammatory agent in ischemia-reperfusion injury. For this, we employed an in vivo rat model with kidney ischemia reperfusion injury to evaluate cardiac injury to clarify the mechanisms of lithium tetraborate. The evaluation of cardiac injury through kidney artery occlusion and reperfusion rat model indicated that lithium tetraborate could (1) reduce oxidative stress-induced endothelial dysfunction; (2) attenuate the inflammatory response of cardiac cells; and (3) alleviate the apoptosis and necrosis of myocytes. In summary, lithium tetraborate demonstrates significant therapeutic properties that contribute to the amelioration of cardiac damage, and it could be a promising candidate for future applications in myocardial dysfunction.

ABSTRACT

Abstract The continuous prolonged exposures of sun light especially the ultra violet (UV) radiation present in it, cause not only the risk of skin cancer but also it may cause premature skin aging, photodermatoses and actinic keratoses. Flavonoids (including Flavane, Flavanone, Flavone, Flavonol, Isoflavone, Neoflavone etc.) having potent antioxidant activity, used as topical applications for protection against UV induced skin damages as well as for skin care. Most commonly used flavonoid is quercetin (Flavonol), which is present in fruits, vegetables, and herbs. We aim to review the research focused on development of different novel formulations to treat UV radiations induced skin diseases. In this review, several formulations of flavonoid quercetin were discussed and their outcomes were compiled and compared in context to solubility, stability and efficiency of application. On the basis this comparative analysis we have concluded that three formulations, namely glycerosomes, nanostructured lipid carriers and deformable liposomes hold good applications for future aspects for topical delivery of quercetin. These formulations showed enhanced stability, increased quercetin accumulation in different skin layers, facilitated drug permeation in skin and long-lasting drug release.

Abstract Hepatoprotective effects of many herbal agents have been reported in animal studies and clinical trials. In this study, five hepatoprotective plants with potent antioxidant, anti-inflammatory, and hypolipidemic effects were chosen to prepare a polyherbal compound for managing NAFLD. Sixty patients with NAFLD were randomly divided into treatment and control groups (2:1 ratio). Both group were advised to take healthy diet and exercise. The treatment group also received herbal capsules containing 400 mg of the mixture of Anethum graveolens, Citrus aurantium, Cynara scolymus, Portulaca oleracea, and Silybum marianum (2 capsules, thrice daily, for two months). The liver ultrasound and biochemical markers including the serum lipids, liver enzymes, and glucose were evaluated before starting the study and at the end of the treatment. Thirty patients in the treatment group and sixteen patients in the control group completed the study. The herbal compound significantly decreased the serum level of alanine transaminase (ALT), aspartate transaminase (AST), and total cholesterol. Treatment with the herbal compound significantly improved the grade of the fatty liver, but no significant change was found in the control group. In conclusion, the formulated herbal compound appeared to be effective in biochemical improvement and decreasing the grade of the fatty liver in the patients with NAFLD.

ABSTRACT

This study aimed to analyze the antiarthritic activity of ginkgolic acid against the Complete Freund's Adjuvant (CFA)-induced arthritis in rats. Arthritis was induced through an intradermal injection of CFA (0.1 mL) at the right hind footpad of adult Wistar Albino rats. Ginkgolic acid was administered orally at doses of 25 mg/kg and 50 mg/kg, respectively, once daily via gavage for 25 days upon inducing arthritis. Indomethacin was administered orally at a dose of 3 mg/kg twice in a week which served as positive control group. The animals were sacrificed and subjected to biochemical and histopathological analysis upon completion of treatment. Ginkgolic acid was able to reverse the arthritic effect (p < 0.01) induced by CFA in a dose dependent manner. Swelling of paw, thymus and spleen index, serum biomarker levels, and pro-inflammatory cytokines were significantly reduced (p < 0.01) by the acid whereas the antioxidant enzyme activities were remarkably restored. The histopathological findings were in agreement with the biochemical results. The results indicate that the antioxidant and anti-inflammatory properties of ginkgolic acid can be credited to the antiarthritic effects, and it can be promoted as a potential agent for therapeutic use against osteoarthritis

Abstract Citrus fruit production occupies a place of considerable importance in the economy of the world including Pakistan. Tristeza disease caused by Citrus Tristeza Virus (CTV) exists in various forms that may or may not cause symptoms in the plants. The bioactive compounds and antioxidants are naturally present in plants and provide a defense mechanism that is generally accelerated in response to a stress. The objective of the present study was to target and analyze the citrus plants that were CTV positive to observe the changes in the enzymatic and non-enzymatic antioxidants of citrus (Sweet Oranges only). It was observed that in response to CTV infection, both the non-enzymatic antioxidants (total flavonoid, ascorbic acid, phenolic acid) and enzymatic antioxidants (catalase, superoxide dismutase and peroxidase) activities showed an increasing trend overall. The profiling of antioxidants in response to a viral infection may help in the discovery of new biomarkers that can be used as a monitoring tool in disease management.


Resumo As frutas cítricas ocupam um lugar de considerável importância na economia do Paquistão, assim como o resto do mundo. A doença da tristeza causada pelo Vírus da Tristeza dos Citros (CTV) existe em várias formas que podem ou não apresentar sintomas nas plantas. Os compostos bioativos e antioxidantes estão naturalmente presentes nas plantas e fornecem um mecanismo de defesa que é geralmente acelerado em resposta a um estresse. O objetivo do presente estudo foi analisar as alterações causadas pelo CTV nos antioxidantes enzimáticos e não enzimáticos de laranjas doces. Foi observado que, em resposta ao ataque de CTV, os antioxidantes não enzimáticos como flavonoides totais, ácido ascórbico, ácido fenólico e antioxidantes enzimáticos, como as atividades de catalase, superóxido dismutase e peroxidase, geralmente mostram uma tendência crescente. O perfil de antioxidantes em resposta a um ataque viral pode ajudar na descoberta de novos biomarcadores que podem ser usados ​​como uma ferramenta de monitoramento no gerenciamento de doenças.

Este trabalho propôs o uso do fármaco quelante mesilato de desferroxamina (DFO) como agente adjuvante para estabilização química e microbiológica de formulações. Soluções de ácido ascórbico (AA) 5,0% (p/v) foram preparadas com sistemas antioxidantes constituídos por diferentes combinações de DFO, ácido etilenodiamino tetra-acético (EDTA) e metabissulfito de sódio, cada adjuvante na concentração máxima de 0,1% (p/v). Os sistemas foram testados previamente quanto à atividade antioxidante, mediante adição de um complexo de ferro (III) redox-ativo e ensaio baseado em fluorescência. Os sistemas também foram associados ao metilparabeno e avaliados quanto à atividade antimicrobiana pelo método turbidimétrico, utilizando-se a técnica de microdiluição em meios líquidos e cepas padrão de bactérias e fungos, incluindo S. aureus (ATCC 6538), E. coli (ATCC 8739), P. aeruginosa (ATCC 9027), C. albicans (ATCC 10231) e A. brasiliensis (ATCC 16404). As soluções de AA foram expostas a condições de teste de estabilidade acelerada e avaliadas quanto à estabilidade química, empregando-se método volumétrico validado para quantificar AA. Verificou-se que o EDTA foi o agente quelante que melhor contribuiu na estabilidade química da solução de AA, entretanto, o DFO apresentou desempenho muito superior ao EDTA para bloquear a atividade pró-oxidante do ferro. Além disso, o DFO foi fator relevante na inibição do crescimento microbiano e demonstrou sinergia com o metilparabeno. A otimização estatística dos resultados indicou que o uso do DFO nos sistemas antioxidante e conservante pode reduzir consideravelmente a concentração dos adjuvantes convencionais, EDTA, metabissulfito e metilparabeno, os quais são muitas vezes associados a reações de hipersensibilidade ou a danos ao meio ambiente


In this work it was proposed the use of the chelating drug desferroxamine mesylate (DFO) as adjuvant for chemical and microbiological stabilization of formulations. Ascorbic acid (AA) solutions 5.0% (w/v) were prepared with antioxidant systems containing different combinations of DFO, ethylenediaminetetraacetic acid (EDTA) and sodium metabisulphite, using a maximum concentration of 0.1% (w/v) for each adjuvant. Previously, the systems were spiked with a redox-active iron (III) complex and tested for antioxidant activity by fluorescence-based assay. The systems also were associated with methylparaben and evaluated for antimicrobial activity by turbidimetric method, using the microdilution technique and standard strains of bacteria and fungi, including S. aureus (ATCC 6538), E. coli (ATCC 8739), P. aeruginosa (ATCC 9027), C. albicans (ATCC 10231) and A. brasiliensis (ATCC 16404). The AA solutions were exposed to accelerated stability test conditions and evaluated for chemical stability, using a volumetric method that was validated to quantify AA. It was found that EDTA was the chelating agent that most contributed to the chemical stability of AA solution, however, DFO demonstrated a much higher performance to EDTA to block the pro-oxidant activity of iron. In addition, DFO was a relevant factor in the inhibition of microbial growth and showed synergy with methylparaben. The statistical optimization of the results indicated that the use of DFO in the antioxidant and preservative systems might considerably reduce the concentration of the conventional adjuvants, EDTA, metabisulphite and methylparaben, which are often associated with hypersensitivity reactions or environmental damage

ABSTRACT The development and validation of a simple and efficient method for the quantification of ferulic acid in poly (D,L-lactide-co-glycolide) (PLGA) nanoparticles coated with chitosan (CS) by reverse phase high performance liquid chromatography coupled to photodiode array detection was described. For the chromatographic analysis, a reverse phase C-18 column was used, mobile phase consisting of acetonitrile and 0.5% acetic acid (37:63, v/v), isocratically eluted at a flow rate of 1 mL/min. Drug determination was performed at 320 nm. The method was validated in terms of the selectivity, linearity, precision, accuracy, robustness, limits of detection and quantification. The method was linear in the range of 10 to 100 µg/mL (r=0.999) and presented limit of detection and quantification of 102 ng/mL and 310 ng/mL, respectively. The method was precise (intra and inter-day) based on relative standard deviation values (less than 3.20%). The recovery was between 101.06 and 102.10%. Robustness was demonstrated considering change in mobile phase proportion. Specificity assay showed no interference from the components of nanoparticles or from the degradation products derived from acidic and oxidative conditions. The proposed method was suitable to be applied in determining the encapsulation efficiency of ferulic acid in PLGA-CS nanoparticles and can be employed as stability indicating one.

Calendula is used widely in cosmetic formulations that present phenolic compounds in their chemical constitution. The objective of our research was to develop and evaluate the stability of topical formulations containing 5% hydro-ethanolic extract of calendula leaves, including spreadability, and in vitro photo-protective, and antioxidant capacity. To evaluate the stability, we used organoleptic characteristics, pH, and viscosity parameters. Antioxidant capacity was measured by the DPPH (2,2-diphenyl-1-picrylhydrazyl) method, and the photo-protective capacity by SPF spectrophotometric measure. All formulations were stable. The calendula extract formulations in gel and cream showed no significant variations in pH, and the cream formulations presented lower viscosity variations than gel formulations. The spreadability of the gel formulations was superior to those in cream. The formulations also presented good antioxidant capacities and an FPS of around 1.75. In accordance with the results, the formulations can be used as antioxidants, but considering the low SPF obtained, calendula cannot be considered as a stand-alone sunscreen, yet may well be tested in future studies towards verifying enhancement of synthetic sunscreens.


A calêndula é amplamente utilizada em formulações cosméticas, apresentando compostos fenólicos em sua constituição química. Desta forma, o objetivo desta pesquisa foi desenvolver e avaliar a estabilidade de formulações tópicas contendo 5% de extrato hidroetanólico das folhas de calêndula, bem como a espalhabilidade, capacidade antioxidante e fotoprotetora in vitro nas mesmas. Para a avaliação da estabilidade, foram usados parâmetros como a verificação das características organolépticas, pH e viscosidade. A capacidade antioxidante foi verificada pelo método do DPPH (2,2-difenil,1- picril-hidrazila) e a capacidade fotoprotetora pela medida espectrofotométrica do FPS. Para as formulações testadas, observou-se que apresentaram uma boa estabilidade. As formulações de creme e gel com extrato de calêndula não apresentaram variações significativas nos valores de pH e o creme apresentou as menores variações de viscosidade em relação ao gel. A espalhabilidade das formulações de gel foi superior à do creme. As formulações também apresentaram uma boa capacidade antioxidante e um FPS em torno de 1.75. De acordo com os resultados, a formulação pode ser utilizada com ação antioxidante, porém com o FPS obtido, a calêndula não pode ser considerada um filtro solar isolado, mas poderá ser testada em estudos futuros para verificar a potencialização de filtros solares sintéticos.

ABSTRACT

Lycopene, a carotenoid and potent antioxidant is found in large quantities in tomatoes. Lycopene combats diseases, such as cardiovascular disease and different types of cancer, including prostate cancer. However, its topical use in emulsion form for the combat of skin aging is under-explored. The aim of the present study was to develop an emulsion containing lycopene extracted from salad tomatoes and evaluate its cytotoxicity, stability, rheological behavior, antioxidant activity and phytocosmetic permeation. The developed cosmetic comprised an oil phase made up of shea derivatives and was evaluated in terms of its physiochemical stability, spreadability, thermal analysis, rheological behavior, microbiological quality, cytotoxicity, antioxidant activity, cutaneous permeation and retention. The results demonstrate that this phytocosmetic is stable, exhibits satisfactory rheological behavior for a topical formula and is a promising product for combating skin aging.


Licopeno é um carotenóide com potente atividade antioxidante encontrado em grande quantidade no tomate e usado no combate a diversas doenças como doenças cardiovasculares e diferentes tipos de cânceres, incluindo o câncer de próstata. O objetivo desse trabalho foi desenvolver uma emulsão contendo extrato de licopeno obtido do tomate salada e avaliar a citotoxicidade do extrato, a estabilidade, o comportamento reológico, atividade antioxidante e permeação do fitocosmético. O cosmético foi desenvolvido utilizando fase oleosa contendo derivados de Karité e submetido à avaliação da estabilidade físico-química, espalhabilidade, análise térmica, comportamento reológico, qualidade microbiológica, citotoxicidade, atividade antioxidante e testes de permeação e retenção cutânea. Os resultados demonstraram que o fitocosmético é estável, apresenta comportamento reológico desejável para uma formulação tópica e é um produto promissor para ser utilizado no combate à aceleração do envelhecimento cutâneo.

RESUMO As úlceras pépticas são decorrentes de um desequilíbrio entre os agentes agressores endógenos, exógenos e agentes protetores. Neste estudo avaliou-se a atividade antiulcerogênica in vivo para o extrato aquoso de Salviaofficinalis (EAS). Após os tratamentos (n = 6; v.o) com o controle (C) água; pantoprazol (30 mg/kg) e EAS (100, 250 e 400 mg/kg, respectivamente) os ratos receberam uma solução de HCl 0,3 M em etanol 60% (v.o) e realizaram-se avaliações morfológicas dos estômagos por meio de uma tabela de escores de lesão (ANOVA; one way e Tukey). EAS (400 mg/kg) apresentou atividade antiulcerogênica (31,47%) semelhante ao pantoprazol (33,83%) em comparação (p<0,0001) com o grupo C. A análise fitoquímica por cromatografia líquida de alta eficiência revelou uma elevada concentração de ácido rosmarínico (3,53%) para EAS. Os flavonoides (0,25%) e os fenóis totais (309,0 EAG) foram quantificados por espectrofotometria de UV/Vis. EAS e ácido rosmarínico apresentaram elevadas atividades antioxidantes por DPPH (Concentração Efetiva 50%-CE50 9,2 e 0,7 mg/mL, respectivamente) justificando pelo menos em parte, o efeito antiulcerogênico do extrato.


ABSTRACT Peptic ulcers are caused by an unbalance among endogenous, exogenous damaging agents and protective agents. In this study we evaluated the in vivo antiulcerogenic activity for the aqueous extract of Salvia officinalis (EAS). After treatments (n = 6; v.o) with control (C) water; pantoprazole (30 mg/kg) and EAS (100, 250 and 400 mg/kg, respectively) the rats received a 0.3 M HCl solution in ethanol 60% (v.o) and it were performed morphological evaluations of the stomachs through the use of a table of injury scores (ANOVA, one way and Tukey). The EAS (400 mg/kg) showed an antiulcerogenic activity (32.53%). Similar to the pantoprazole (29.91%), in comparison with (p<0.0001) the group C. The phytochemical analysis through high performance liquid chromatography revealed high acid rosmarinic (3.53%) for the EAS. The flavonoids (0.25%) and the total phenolics (GAE 309.0) were quantified by spectrophotometry UV/Vis. The EAS and the rosmarinic acid exhibited elevated antioxidant activities by DPPH (Effective Concentration 50% -EC50 9.2 and 0.7 mg/mL, respectively,) at least partially explaining the antiulcerogenic effect of the extract.

ABSTRACT

Free radicals produced in the body can be explained by several factors and induce cell damage in case of oxidative stress. In order to fight them, there are the endogenous antioxidant defense mechanisms, or exogenous, when derived from the diet. Some vitamins and minerals, besides phenolic compounds, present antioxidant function and fight these radicals. Therefore, many studies have demonstrated an association between the consumption of food with antioxidants and health benefits. Assessing the intake of antioxidants provides intervention measures and public policy actions. Several different instruments can be used to assess this intake, but each of them should be well known in order to make the most appropriate choice. Therefore, the aim of this study is to approach the different types of diet surveys used to determine the food intake of antioxidants, as well as discuss their advantages and limitations of use. The following surveys were used to determine the intake of antioxidants: Dietary Records, 24-hour recall, Food Frequency Questionnaire, and Antioxidant Food Frequency Questionnaire. Most of the surveys meth¬ods used consider only the antioxidant micronutrients, but one of the them quantifies the total antioxidant capacity of the diet, including the phenolic compounds. The methods analyzed present advantages and disadvantages, in addition to those inherent to the instrument. Critical reflection about the choice of the method allows a better assessment of the intake of antioxidants, aiding in a more accurate food diagnosis, eventually ensuring food and nutritional security for the population through necessary public policy initiatives.


Los radicales libres producidos en el organismo pueden explicarse por varios factores y causar daños celulares en el caso de que aparezca estrés oxidativo. Para combatirlos existen los mecanismos de defensa antioxidantes endógenos o exógenos provenientes de la dieta. Algunas vitaminas y minerales, además de los compuestos fenólicos, son los responsables de la función antioxidante y de combatir los radicales. Por lo tanto, muchos estudios han asociado el consumo de alimentos ricos en antioxidantes como algo beneficioso para la salud. Para evaluar la ingesta de antioxidantes se necesitan medidas de intervención y acciones de política pública. Para evaluar la ingesta se pueden utilizar diferentes instrumentos, sin embargo, se debe conocer cada uno de ellos para hacer la elección correcta. Por lo tanto, el objetivo de este estudio es abordar los diferentes tipos de encuestas alimentarias utilizadas para determinar el consumo de antioxidantes, así como discutir las ventajas y límites de su uso. Las diferentes encuestas utilizadas para determinar el consumo de antioxidantes fueron Registro Alimentar, Recordatorio 24 horas, Cuestionario de Frecuencia Alimentaria y Cuestionario de Frecuencia Alimentaria especifico para antioxidantes. La mayoría de los análisis se hace considerando sólo micronutrientes antioxidantes, sin embargo, en uno de los métodos, se cuantifica la capacidad antioxidante total de la dieta, incluyendo los compuestos fenólicos. Las metodologías abordadas presentan ventajas e inconvenientes, además de las inherentes al instrumento. La reflexión para la elección crítica del método permite una mejor evaluación de la ingesta de antioxidantes, ayudando en la precisión del diagnóstico alimentario. Y, por último, se garantiza la seguridad alimentaria y nutricional de la población por medio de iniciativas de políticas públicas necesarias.


Os radicais livres produzidos no organismo podem ser explicados por fatores diversos e induzem a danos celulares caso ocorra estresse oxidativo. A fim de combatê-los, existem os mecanismos endógenos de defesa antioxidante ou exógenos, quando provenientes da dieta. Algumas vitaminas e minerais, além de compostos fenólicos, são responsáveis por apresentar função antioxidante e combater os radicais. Por isso, muitos estudos vêm associando o consumo de alimentos fonte de antioxidantes aos benefícios à saúde. Avaliar a ingestão de antioxidantes propicia medidas de intervenção e ações de políticas públicas. Para avaliação da ingestão, podem ser utilizados diferentes instrumentos, porém deve-se conhecer cada um deles para realizar a escolha adequada. Sendo assim, o objetivo deste estudo é abordar os diferentes tipos de inquérito alimentar utilizados para determinar a ingestão de antioxidantes, assim como discutir suas vantagens e limitação de uso. Os diferentes inquéritos utilizados para determinar a ingestão de antioxidantes foram Registro Alimentar, Recordatório 24 horas, Questionário de Frequência Alimentar e Questionário de Frequência Alimentar específico para antioxidantes. A maioria realiza análise considerando apenas os micronutrientes anti-oxidantes, porém, em um dos métodos, quantifica-se a capacidade antioxidante total da dieta, incluindo-se também os compostos fenólicos. As metodologias abordadas apresentam vantagens e desvantagens, além daquelas inerentes ao instrumento. A reflexão para a escolha crítica do método permite melhor avaliação da ingestão de antioxidantes, auxiliando no diagnóstico alimentar mais preciso. Por fim, garante a Segurança Alimentar e Nutricional da população por meio de iniciativas de políticas públicas necessárias.

ABSTRACT

Phenolic compounds or polyphenols are bioactive compounds widely distributed in many foods such as fruits, vegetables, tea and wine. Several studies indicate the important role of these compounds in the prevention of chronic diseases such as cancer and cardiovascular diseases. Furthermore, polyphenols may be considered pharmacological agents capable of combating oxidative stress owing to reduced formation of reactive oxygen species and inflammation. The imbalance between the production of free radicals and antioxidant capacity is a common feature in patients with chronic kidney disease (CKD) and is associated with progression of disease, and is considered a risk factor for cardiovascular disease, the leading cause of mortality in these patients. Although scarce, some studies have shown the benefits of using phenolic compounds in patients with chronic kidney disease and, therefore, the present review discusses the possible applicability of phenolic compounds in this population.


Los compuestos fenólicos o polifenoles son compuestos bioactivos ampliamente distribuidos en muchos alimentos, tales como frutas, legumbres, verduras, tés y vino. Varios estudios indican el importante papel de estos compuestos en la prevención de enfermedades crónicas, tales como cáncer y enfermedades cardiovasculares. Además, debido a sus propiedades anti-inflamatorias y antioxidantes, los polifenoles pueden ser considerados agentes farmacológicos capaces de combatir el estrés oxidativo, debido a la reducción de la formación de especies reactivas del oxígeno o radicales libres. El desequilibrio entre la producción de radicales libres y la capacidad antioxidante es una característica común de los pacientes con enfermedad renal crónica (DRC) y está relacionado con la progresión de la lesión renal, además de que se considera un factor de riesgo para las enfermedades cardiovasculares, la principal causa de mortalidad en estos pacientes. Aunque sean escasos, algunos estudios han demostrado los beneficios de la utilización de compuestos fenólicos en los pacientes con DRC y, por lo tanto, la presente revisión pretende debatir la posible aplicabilidad de los compuestos fenólicos en esta población.


Os compostos fenólicos ou polifenóis são compostos bioativos amplamente distribuídos em diversos alimentos, como frutas, legumes, verduras, chás e vinho. Diversos estudos apontam o importante papel destes compostos na prevenção de doenças crônicas, como câncer e doenças cardiovasculares. Além disso, em virtude de suas habilidades anti-inflamatórias e antioxidantes, os polifenóis podem ser considerados agentes farmacológicos capazes de combater o estresse oxidativo devido à redução na formação de espécies reativas de oxigênio ou radicais livres. O desequilíbrio entre a produção de radicais livres e a capacidade antioxidante é uma característica comum nos pacientes com doença renal crônica (DRC) e está associado à progressão da lesão renal, além de ser considerado um fator de risco para doenças cardiovasculares, a principal causa de mortalidade nestes pacientes. Apesar de escassos, alguns trabalhos têm mostrado os benefícios do uso dos compostos fenólicos nos pacientes com DRC e, portanto, a presente revisão pretende discutir a possível aplicabilidade dos compostos fenólicos nesta população.

ABSTRACT

Serjania erecta Radlk.(Sapindaceae) is a medicinal plant traditionally used in Brazil. We assayed the ethanolic extract of leaves and roots against seven microorganisms. The REMA (Resazurin Microtiter Assay) assay was used to measure the biological activity in vitro against Mycobacterium tuberculosis and our results showed moderate activity of the ethanolic extract. On the other hand S. aureus, P. aeruginosa, S. setubal, C. albicans, S. cerevisiae and E. coli revealed that the leaves and roots of S. erecta inhibited the growth of all microorganisms. The ethanolic extracts of leaves and roots showed low values of antioxidant activities. The ethanolic extracts of leaves and roots were analyzed by chromatographic and spectrometric methods. (-)-Epicatechin, kaempferol aglycone and five glycoside derivates were isolated: kaempferol-3-O-α-L-rhamnopyranoside, kaempferol-3-O-α-L-rhamnopyranosyl-(1→6)-β-D-glucopyranoside from the roots and kaempferol, kaempferol 3,7-di-O-α-L-rhamnopyranoside, vitexin, isovitexin and (-)-epicatechin in the leaves. This is the first chemical study reported in the literature about this specie.


Serjania erecta Radlk. (Sapindaceae) é uma planta medicinal utilizada no Brasil. Analisamos o extrato etanólico de folhas e raízes para sete microorganismos. O REMA (Ensaio de Microtitulação Resazurina) foi utilizado para medir a atividade biológica in vitro contra o Mycobacterium tuberculosis e nossos resultados mostraram atividade moderada do extrato etanólico. Por outro lado, ensaios com S. aureus, P. aeruginosa, S. setubal, C. albicans, S. cerevisiae e E. coli revelaram que as folhas e raízes de S. erecta inibiram crescimento em todos os microorganismos. Os extratos etanólicos de folhas e raízes apresentaram valores baixos de atividade antioxidante. Os extratos etanólicos de folhas e raízes foram analisados por métodos cromatográficos e espectroscópicos. (-)-Epicatequina, canferol aglicona e cinco derivados glicosídicos foram isolados: canferol -3-O-α-L-ramnopiranosídeo, canferol-3-O-α-L-ramnopiranosil-(1→6)-β-D-glucopiranosídeo a partir das raízes e canferol, canferol 3,7-di-O-α-L-ramnopiranosídeo, vitexina, isovitexina e (-) epicatequina nas folhas. Este é o primeiro estudo químico na literatura sobre esta espécie.

ABSTRACT

O vinho tinto é rico em compostos fenólicos com atividade antioxidante, capazes de inativar espécies reativas de oxigênio, minimizando danos celulares oriundos do estresse oxidativo, proporcionando uma redução de risco para doenças crônicas não transmissíveis. Assim, os objetivos desta pesquisa foram identificar associações entre a atividade antioxidante in vitro e fatores relacionados ao tipo de uva, região de produção, perfil sensorial, safra, valor comercial e concentração de compostos fenólicos de vinhos tintos produzidos no Brasil, Chile e Argentina. Inicialmente, os vinhos brasileiros (n=29) foram avaliados em relação à atividade antioxidante (ORAC e DPPH), cor instrumental e compostos fenólicos majoritários, no intuito de verificar qual classe de fenólicos estaria associada com a atividade antioxidante. Verificou-se que tanto os compostos fenólicos totais como os flavonóides totais, com destaque aos flavonóides não-antociânicos, se associaram significativamente (p<0,05) com a atividade antioxidante. Em um segundo passo, as características sensoriais, a cor, o valor comercial e a atividade antioxidante das 80 amostras de vinhos Sul-Americanos, distribuídas em Merlot (n=9), Pinot Noir (n=17), Malbec (n=11), Syrah (n=12), Cabernet Sauvignon (n=24), e vinhos de uvas americanas (Vitis labrusca) (n=7) foram avaliados usando estatística multivariada, objetivando-se verificar se a qualidade sensorial das amostras estaria associada com o valor comercial, cor, e à atividade antioxidante. De uma forma geral, os vinhos chilenos e argentinos apresentaram maior atividade antioxidante, valor comercial, intensidade de odor, qualidade sensorial, índice de acidez e taninos, ao passo que os vinhos brasileiros obtiveram os menores valores para os atributos sensoriais. Os vinhos de uvas americanas apresentaram menores valores para todas as variáveis. As varietais Syrah, Malbec e Cabernet Sauvignon apresentaram maior capacidade antioxidante e melhores características ...


Red wine is rich in phenolic compounds with antioxidant activity, being able to buffer reactive oxygen species, thus decreasing the risk of non-transmissible chronic diseases. In this regard, the objectives of this research aimed at identifying associations between the in vitro antioxidant activity and factors related to grape varietal, region of production, sensory profile, vintage, color, commercial value, and concentration of phenolic compounds of red wines produced in Brazil, Chile, and Argentina. Initially, the Brazilian red wines (n = 29) were assessed in relation to antioxidant activity, instrumental color, and major phenolic compounds with the objective to verify which phenolic class was associated with the antioxidant activity. Both the total phenolic compounds and total flavonoids, with special attention to non-anthocyanin flavonoids, were significantly associated with the antioxidant activity. In a second step, the sensory characteristics, color, commercial value, and antioxidant activity of the 80 red wine samples, which were distributed in Merlot (n=9), Pinot Noir (n=17), Malbec (n=11), Syrah (n=12), Cabernet Sauvignon (n=24), and table wines (Vitis labrusca) (n=7) were evaluated using multivariate statistical techniques, with the aim to verify how the overall perception of quality of wines was related to commercial value, color and antioxidant activity. In a general way, te Chilean and Argentinean red wines displayed a higher antioxidant activity, commercial value, intensity of odors, sensory perception of quality, acidity level, and tannin level, whereas the Brazilian samples obtained the lowest values for the sensory attributes. The table wines presented the lowest values for all response variables. Syrah, Malbec and Cabernet Sauvignon varietals presented the highest antioxidant activity and most favorable sensory features, and this result was independent of wine's vintage and origin. ABSTRACT

The association between high levels of LDL cholesterol and oxidative stressis the on set of the atherosclerotic process. Oxidative stress, verified by the significant increase in the concentration of reactive oxygen species, causes damage to the endothelial wall and leads to LDL oxidation, which in turn will be deposited as fatty cells leading to the obstruction of vessels and arteries. In vascular cells a lot of enzymes have been described as potential sources of ROS; however, enzymatic and non enzymatic mecanisms protect the organism against such species. Among non enzymatic antioxidants there are substances derived from foods such as vitamins and phenolic compounds, which have been receiving increasing attentiondue to their chemopreventive function against oxidative damages. Rosemary (Rosmarinus offi cinalis L.) belongs to the Labiatae family, the antioxidant properties of which have been attributed to a variety of phenolic compounds capable of reacting with free radicals and quench ROS. Studies have revealed that rosemary could be an important compound in antiglycative therapy, helping in the prevention and treatment of chronic-degenerative diseases, reinforcing the need of new researches to elucidate its antioxidant contribution in vivo.


La asociación entre niveles elevados de colesterol LDL y el estrés oxidativo compone el inicio del proceso arteriosclerótico y contribuye para el desarrollo de enfermedades cardiovasculares. El estrés oxidativo, determinado por el aumento significativo de la concentración de especies reactivas del oxígeno (EROS), produce daños en la pared endotelial y oxidación de las LDL queserán depositadas como células grasas provocando la obstrucción de vasos y arterias. En las células vasculares muchas enzimas hansido descritas como fuentes potenciales de EROS, pero mecanismos enzimáticos y no enzimáticos protegen el organismo contra esas especiesreactivas. Entre los antioxidantes no enzimáticos hay sustancias derivadas de los alimentos comolas vitaminas y los compuestos fenólicos, los cuales han recibido una gran atención debidoa sus propiedades quimio preventivas contra el daño oxidativo. El romero (Rosmarinus offi cinalisL.) pertenece a la familia Labiatae cuyas propiedades antioxidantes han sido atribuidasa una variedad de compuestos fenólicos capacesde reaccionar con los radicales libres y eliminarlas EROS. Estudios han revelado que el romero puede ser un elemento importante en el combatea la oxidación glicativa, auxiliando en la prevención y tratamiento de las enfermedades degenerativas crónicas y reforzando la necesidadde aumentar las investigaciones para dilucidarsu contribución antioxidante in vivo.


A associação entre níveis elevados de LDL colesterol e o estresse oxidativo constituem o início do processo aterosclerótico e contribuem para o desenvolvimento das doenças cardiovasculares. O estresse oxidativo, verifica do pelo aumento significativo da concentração de espécies reativas de oxigênio (EROS), causa danos na parede endotelial e leva à oxidação das partículas de LDL, que por sua vez serão depositadas como células gordurosas levando à obstrução dos vasos e artérias. Nas células vasculares, muitas enzimas têm sido descritas como fontes potenciais de EROS, contudo mecanismos enzimáticos e não-enzimáticos protegem o organismo contra tais espécies reativas. Dentre os antioxidantes não enzimáticos encontram-se substâncias derivadas de alimentos como vitaminas e compostos fenólicos, os quais têm recebido crescente atenção devido às suas funções quimio preventivas contra o dano oxidativo. O alecrim (Rosmarinus officinalis L.) é um membro da família Labiatae, cujas propriedades antioxidantes têm sido atribuídas a uma variedade de compostos fenólicos capazes de reagir com radicais livres e eliminar as EROS. Estudos relatam que o alecrim pode ser um componente importante na terapia antiglicativa, auxiliando na prevenção e no tratamento de doenças crônico-degenerativas, reforçando a necessidade de novas pesquisas para elucidar sua contribuição antioxidante in vivo.

Este estudo foi estruturado em cinco capítulos. No capítulo I temos um breve referencial teórico sobre a importância da carne de frango, os mecanismos da oxidação lipídica e a utilização de antioxidantes naturais. O capítulo II traz os ensaios da avaliação da atividade antioxidante in vitro do mel e das especiarias orégano (Origanum vulgare L.) e sálvia (Salvia officinalis L.) durante a vida de prateleira. Os resultados de fenólicos totais do orégano indicaram um aumento de 1154,09 a 1611,28 mgEAG/mL (O a 12 meses) e na sálvia os valores variaram entre 1309,8 a 2032,4 mgEAG/mL no decorrer do tempo (O a 12 meses). Os resultados da porcentagem da inibição da oxidação lipídica (% IOL), pelo sistemas β-caroteno/ácido linoléico mostrou que a sálvia inibiu a oxidação em 74,6, 81,3 e 81,3%, nos tempos (0,6 e 12 meses) e o orégano apresentou valores de inibição de (43,2, 63,3 e 50,7%). Quando se avaliou o índice de atividade antioxidante (IAA) utilizando o aparelho Rancimat, a sálvia apresentou um índice de atividade antioxidante (3,35) superior aos demais, que apresentaram 1,69, 1,25 e 1,08 para o BHT, orégano e mel, respectivamente. Os resultados do ensaio da capacidade de absorbância do radical oxigênio (ORAC) revelou que o orégano apresentou valores de 544,6, 430,7 e 1019,6 ET µmol/g, nos tempos O, 6 e 12 meses, respectivamente...


This study has been structured into five chapters. Chapter I provides a brief historical theoretical reference point regarding the importance of chicken meat, the mechanisms of lipid oxidation and the use of natural antioxidants. Chapter II presents trials evaluating the in vitro antioxidant activity, of honey and the spices oregano (Origanum vulgare L.) and sage (Salvia officinalis L.) over their shelf life. The results for oregano indicated an increase in total phenols from 1154.09 to 1611.28 mg of gallic acid equivalent (GAE)/100 g (0 to 12 months). For sage, the values changed from 1309.8 to 2032.4 mg GAE/100 g over the period (0 to 12 months) and for honey, the values measured were 1007.1, 1830.4 and 2129.9 mg GAE/100 g at the times of 0,6 and 12 months, respectively. The results relating to the percentage inhibition of lipid oxidation(% ILO) by the ß-carotene/linoleic acid system showed that sage inhibited oxidation by 74.6, 81.3 and 81.3% at the times of 0,6 and 12 months, while oregano presented inhibition values of 43.2, 63.3 and 50.7%. Evaluation of the antioxidant activity index (AAI) using the Rancimat apparatus showed that the AAI for sage (3.35) was greater than the indices for the other agents, which were 1.69, 1.25 and 1.08 for BHT, oregano and honey, respectively. The results from testing the oxygen radical absorbance capacity (ORAC) showed that oregano presented values of 544.6, 430.7 and 1019.6 TE µmol/g at the times of 0,6 and 12 months, respectively. Sage presented ORAC values of 610.45, 467.44 and 822.21 at the times of 0,6 and 12 months and honey presented 47.3, 22.4 and 26.1 ET µmol/g, at the times of 0,6 and 12 months. Comparing these results with those described in the literature, it can be concluded that these herbs and honey have high potential as antioxidants. Chapter III shows the influence of the bioactive compounds in sage and oregano for protection against lipid oxidation, in a microsomal substrate of chicken meat. The mean concentrations of TBARS (µmol of MDA/mg of protein) observed in the breast meat samples were, for the control, 7.45; for BHT, 1.91; and for oregano+sage, 3.45. In the thigh meat samples, the following results were observed: control (9.83); BHT (4.27); oregano+sage (3.15). The treatments (BHT and oregano+sage) reached their peak inhibition after three hours (82.42% and 82.25%, respectively). However, analysis of the inhibition of lipid oxidation in the microsomal fraction of the thigh meat showed that the BHT treatment reached its peak inhibition (66.50%) after one hour of induction and the oregano+sage treatment reached its peak inhibition after three hours (82.25%). The results relating to inhibition of lipid oxidation during the induction period showed that, in relation to the control, the treatments implemented had a positive influence regarding protection against lipid oxidation in a microsomal substrate of chicken breast meat. Chapter IV evaluates the effect of spices and honey with regard to protecting against lipid oxidation in a system consisting of a homogenate model of chilled chicken meat. The results from water action on the breast and thigh meat homogenates (either, raw or cooked) showed that the treatment (oregano+sage+10%honey) reduced the quantity of free water over the period of refrigeration. With regard to the pH values in the breast meat homogenates, it was seen that they rose over the period of refrigeration, in all the treatments evaluated. In the thigh meat, the pH values were higher than those observed in the breast meat. In the homogenates of raw breast meat, a marked loss of moisture was observed in all of the treatments and, particularly, at all durations of refrigeration. In the samples of cooked breast meat, no significant differences in moisture were seen between the control and the treatment with oreganó+sage+5%honey. In the homogenates of raw thigh meat, the moisture values ranged from 60.82 to 66.96 g/100 g. The myoglobin content in the homogenates of raw breast meat ranged from 1.95% to 2.01% at time zero. ABSTRACT

O azeite de oliva é um dos principais ingridientes da Dieta do Mediterâneo, que apresenta relação inversa com a doença aterosclerótica. O efeito cardioprotetor dessa dieta tem sido atribuído em grande parte, à formulação exclusiva do azeite de oliva virgem. O alto teor de ácidos graxos monoinsaturados e de compostos fenólicos, especialemnte oleuropeína e o hidroxitirosol, está relacionado a ações antiaterogênicas, como redução da colesterolemia, redução da agregação plaquetária e diminuição da expressão de moléculas de adesão. Não existe grande variação no teor de ácidos graxos monoinsaturados no que se refere à qualidade de azeite, contudo a maior concentração de compostos fenólicos está no azeite extravirgem, obtido a partir de todo o fruto por meio da primeira prensa física a frio.
